Abstract
|
|
|
Background:
The measurement of the circulating levels of estradiol (E2), progesterone (P4), and luteinizing hormon (LH) is essential for assessing ovarian function and monitoring follicular development during ovarian hyperstimulation in vitro
fertilization
(IVF) procedures. The most widely used method for the determination of these hormones is radioimmunoassay (RIA), but its radiohazard and relatively short half-life of isotopes necessitate alternative methods. Recently a new assay for the
determination
of hormones became available on the automated chemiluminescence immunoassay(CIA), ACS:180TM (Ciba-Corning, USA). We evaluated this assay in the aspects of precision, recovery rates and correlation with RIA.
Methods:
Serum samples taken from patients undergoing gonadotrophin stimulation on IVF program were tested concurrently for E2, P4 and LH by CIA and RIA.
Results:
We got good precision results with control sera. The within-and between-assay precision revealed less than 10%(C.V). The recovery rates of E2, P4 and LH were 83%, 85% and 118%, respectively. Sensitivities for E2, P4 and LH were 13.1 pg/ml,
0.25
ng/ml and 0.2 mIU/ml, respectively. The assay correlated well with RIA (E2:n=21, RIA=0.97¡¿CIA-16.55, r=0.922, P4;n=8, RIA=2.02¡¿CIA-13.57, r=0.986, LH;n=10, RIA=0.50¡¿CIA+0.28, R=0.975)
Conclusions:
ACS : 180TM has an excellent precision, a high digree of sensitivity and a good stability. The assay offer a rapid determination of E2, P4 and LH concentrations, which may be particularly useful for monitoring the menstrual cycle and ovarian
hyperstimulation.
